In order to solve the dependence on petroleum resources, we need to develop new renewable fuel energy. Fatty acid biosynthesis system, a route for producing new biofuels, recently has received significant attention. In order to further understand the fatty acid biosynthesis system, we reconstituted the system in vitro. In the in vitro reconstituted system, we not only further assessed the influence of the concentration of coenzyme and substrate on the system, but also measured the contribution of each component on the entire system. We optimized the fatty acid synthesis system according to the obtained information, and the fatty acid synthesis rate was significantly improved. This system provides a new platform for the future research of fatty acid synthesis, and can effectively guide the industrialization of new bioenergy. In other way, we created the new pathway for fatty alcohols overproduction and combined the fatty acids and 2-keto acids pathway to build new type biodiesel. Furthermore, we have successfully transferred this targeted engineering strategy into terpenoids overproduction.
